From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Cardiac Troponin T in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

Calibrated across a product range of 15.6–1000 pg/ml with a stated sensitivity of <5.5 pg/ml, the assay is read colorimetrically at 450 nm on a microplate reader. The pre-coated 96-well microplate is configured as 12 × 8 well strips, and the total assay time is 4 h 30 min. Reported recovery values are: serum 80%–102% (average 91%, n=5), EDTA plasma 81%–100% (average 90%, n=5) and heparin plasma 80%–89% (average 84%, n=5). Linearity upon dilution (1:2, 1:4, 1:8, 1:16) is also reported for serum, EDTA plasma and heparin plasma samples.

In the sandwich ELISA workflow, an antibody specific for Cardiac Troponin T is pre-coated onto the plate. Standards and test samples are added; the target in each sample binds to the immobilised antibody. After washing, a Biotinylated Anti-Cardiac Troponin T Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow after addition of stop solution; absorbance at 450 nm is proportional to the amount of Cardiac Troponin T captured, and the concentration is read from the standard curve. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage temperatures specified per component.
