Human Troponin I Type 2, Fast Skeletal ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Troponin I Fast Skeletal Muscle in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for Troponin I Fast Skeletal Muscle. Standards and test samples are added to the wells, where the target is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-Troponin I Fast Skeletal Muscle Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of analyte captured, and concentration is calculated from the optical density read at 450 nm in a microplate reader against a standard curve.

The quantitative sandwich assay is read on a colorimetric microplate reader. The product range is 78.1-5000 pg/ml and sensitivity is <29.2 pg/ml. Total assay time is 4h 30m. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. Included items comprise a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers.

Average recovery values across n=5 samples are 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 is reported as 87-91%, 87-107%, 74-101% and 92-97% for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).
