Human Tropomyosin 3 ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables in vitro quantitative determination of human Tropomyosin 3 in serum, plasma, tissue homogenates or other biological fluids [S1]. The assay uses a microtiter plate pre-coated with an antibody specific for Tropomyosin 3, a Biotinylated Anti-Tropomyosin 3 Antibody, and an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450 nm [S1].

The assay has a measuring range of 0.312–20 ng/ml and a sensitivity of 0.126 ng/ml [S1]. It is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips, takes 4 h 30 min to perform, and is read on a colorimetric microplate reader [S1]. Reported spike-and-recovery values are 91% on average for serum (n=5, range 80%–102%), 90% for EDTA plasma (n=5, range 81%–100%), and 84% for heparin plasma (n=5, range 80%–89%) [S1]. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 sample dilutions is 87–107% for serum, 79–108% for EDTA plasma, and 82–105% for heparin plasma [S1].

Kit components include lyophilised standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), 5 adhesive plate sealers and the pre-coated 96-well microplate [S1]. Components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date [S1].
