Human Triosephosphate Isomerase 1 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of human Triosephosphate Isomerase 1 in serum, plasma or other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells and the immobilised antibody captures target. After washing, a Biotinylated Anti-Triosephosphate Isomerase Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that turns yellow upon addition of stop solution; the density of yellow is proportional to the amount of target captured. Absorbance is read at 450 nm on a colorimetric microplate reader and the concentration calculated by reference to a standard curve.

Key performance characteristics include an assay range of 0.625-40 ng/ml and a sensitivity of <0.244 ng/ml, with a total assay time of 4h 30m. Reported average recovery is 91% (range 80%-102%) for serum (n=5), 90% (range 81%-100%) for EDTA plasma (n=5), and 84% (range 80%-89%) for heparin plasma (n=5). Linearity upon dilution at 1:2, 1:4, 1:8 and 1:16 was 87-91%, 87-107%, 74-101% and 92-97% respectively for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).

Kit components include the pre-coated microplate strips, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, stored at +4°C or -20°C as indicated. The kit should not be used past its expiration date.
