From Antibodies.com, this Human PBR ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human PBR in tissue homogenates, cell lysates, cell culture supernates and other biological fluids, with reactivity confirmed for human samples. The assay uses a pre-coated microplate to which standards and samples are added; PBR present in each sample is captured by an immobilised antibody. After incubation and washing, a biotinylated anti-PBR detection antibody binds the captured target, followed by an HRP-avidin conjugate and TMB substrate. Addition of an acidic stop solution yields a yellow product whose intensity is proportional to PBR concentration; absorbance is read at 450 nm in a microplate reader and quantified against a standard curve.

Reported performance figures include a standard range of 0.312-20 ng/ml and a sensitivity of <0.128 ng/ml. The total assay time is approximately 4h 30m. In spiked recovery testing (n=5), average recoveries were 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%) and 84% in heparin plasma (range 80-89%). Kit components are stored at +4°C or -20°C as indicated on each reagent and the kit should not be used past its expiration date.
