Human Transketolase ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of human Transketolase in serum, plasma, tissue homogenates or other biological fluids.

The assay uses a sandwich enzyme immunoassay format on a pre-coated 96-well microplate. Standards and test samples are added to the wells, where Transketolase is captured by an immobilised specific antibody. After washing, a Biotinylated Anti-Transketolase Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate is then added and visualises the HRP enzymatic reaction as a blue product that turns yellow upon addition of acidic stop solution. The absorbance is read at 450 nm on a colorimetric microplate reader, and the density of the yellow colour is proportional to the Transketolase concentration in each sample.

The assay covers a detection range of 0.156-10 ng/ml with a sensitivity of 0.066 ng/ml, and the total run time is 4h 30m. Recovery in spiked serum (n=5) was 80%-102% (average 91%); in EDTA plasma (n=5) 81%-100% (average 90%); and in heparin plasma (n=5) 80%-89% (average 84%). Kit components are stored at +4°C or -20°C as instructed.
