Description

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of human TRPM1 in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique: an antibody specific for TRPM1 is pre-coated onto a microtiter plate, standards and test samples are added, and following incubation and washing the wells are incubated with Biotinylated Anti-TRPM1 Antibody, followed by HRP-Avidin conjugate and TMB substrate solution. The density of the resulting yellow product is proportional to the amount of TRPM1 captured in each well, read at 450 nm in a colorimetric microplate reader.

Sensitivity is reported as <0.051 ng/ml, with an assay range of 0.156–10 ng/ml. Reported recovery averages are 91% for serum (range 80%–102%, n=5), 90% for EDTA plasma (range 81%–100%, n=5), and 84% for heparin plasma (range 80%–89%, n=5). Linearity data across dilutions of 1:2, 1:4, 1:8, and 1:16 are also provided for serum, EDTA plasma, and heparin plasma samples (n=5 each). Total assay time is 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past the expiration date.

Detection is colourimetric on a pre-coated microplate platform. The kit includes a pre-coated microplate, lyophilized standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and adhesive plate sealers. It is intended for use with human samples.
