Human Trans Golgi Network Protein 2 ELISA Kit

A sandwich ELISA from Antibodies.com for the quantitative in vitro determination of human TGN46 (Trans Golgi Network Protein 2) in tissue homogenates, cell lysates, or other biological fluids. The assay employs a 96-well microtiter plate pre-coated with an antibody specific for TGN46. Standards and samples are added to the wells, and captured TGN46 is detected using a Biotinylated Anti-TGN46 Antibody followed by an HRP-Avidin conjugate. A TMB substrate is used to visualise the reaction, producing a blue colour that changes to yellow after addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated by reference to a standard curve.

The kit is reported with a sensitivity of 0.116 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of approximately 4 h 30 min. Components include a pre-coated 96-well microplate (12 × 8 well strips), two vials of lyophilised standard, Detection Solution A and Detection Solution B, a 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and adhesive plate sealers, stored at +4 °C or −20 °C as instructed. Reported recovery values are 80%–102% (average 91%) for serum, 81%–101% (average 91%) for EDTA plasma, and 80%–89% (average 84%) for heparin plasma.
