Human Topoisomerase II ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Topoisomerase II alpha in tissue homogenates, cell lysates, or other biological fluids.

The assay employs a pre-coated 96-well microplate (12 x 8 well strips) with an antibody specific for Topoisomerase II alpha. Standards and samples are added to the wells, and captured analyte is detected using a biotinylated anti-Topoisomerase II alpha antibody, followed by HRP-avidin conjugate and TMB substrate. The resulting yellow product is read at 450 nm in a microplate reader, with absorbance proportional to analyte concentration. The measuring range is 0.625–40 ng/ml with a sensitivity of <0.225 ng/ml, and the assay is completed in 4 hours 30 minutes.

Recovery was 80%–102% (average 91%) in serum, 81%–100% (average 90%) in EDTA plasma, and 80%–89% (average 84%) in heparin plasma (n=5 each). Dilution linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions in all three sample types. Kit components include a pre-coated 96-well microplate, lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and 5 adhesive plate sealers. Components are stored at +4°C or –20°C as instructed. A colorimetric microplate reader is required.
