A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com for the quantitative in vitro determination of human TIRAP (Toll Interleukin 1 Receptor Domain Containing Adaptor Protein) in tissue homogenates and other biological fluids. The kit is reactive with human samples and uses a pre-coated 96-well microplate configured as 12 x 8 well strips, with colourimetric detection read at 450 nm in a microplate reader. Total assay time is 4 hours 30 minutes.

An antibody specific for TIRAP is immobilised on the plate; standards and samples are added, followed by a biotinylated anti-TIRAP detection antibody and an HRP-Avidin conjugate. TMB substrate is converted to a yellow product whose absorbance is proportional to TIRAP concentration, allowing quantification against a standard curve. The manufacturer-stated assay range is 0.312–20 ng/ml with a sensitivity of <0.109 ng/ml.

Manufacturer validation reports average recovery of 91% (range 80–102%) in serum, 90% (range 81–100%) in EDTA plasma and 84% (range 80–89%) in heparin plasma (n=5 each). Dilution linearity is provided for 1:2, 1:4, 1:8 and 1:16 sample dilutions across the same three matrices. Kit components include the pre-coated plate, lyophilised standard, detection solutions A and B, 30× wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers; storage temperatures vary by component between +4°C and -20°C, and the kit should not be used past its expiration date.
