A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com for the in vitro quantitative determination of human TRAF5 in serum, plasma, tissue homogenates or other biological fluids. The assay uses an antibody specific for TRAF5 pre-coated onto a microtiter plate; standards and test samples are added into the wells and TRAF5 present in each sample is bound to the wells by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-TRAF5 Antibody is added, followed by HRP-Avidin conjugate and further incubation. TMB substrate solution is then used to visualize the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of TRAF5 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.
