Human Tight Junction Protein 1 ELISA Kit

Manufactured by Antibodies.com, this Sandwich ELISA is intended for the in vitro quantitative determination of human Tight Junction Protein 1 (ZO1) in tissue homogenates, cell lysates and other biological fluids. The kit employs the sandwich enzyme immunoassay technique on a 96-well microplate pre-coated with antibody specific for ZO1 Tight Junction Protein. Captured analyte is detected with a Biotinylated Anti-ZO1 Tight Junction Protein Antibody followed by an HRP-Avidin conjugate, and the reaction is visualised using TMB substrate, which produces a yellow product after addition of acidic stop solution. The density of yellow is proportional to the analyte concentration, with absorbance read at 450 nm on a colorimetric microplate reader.

Reported performance characteristics include a product range of 0.156–10 ng/ml and a sensitivity of <0.068 ng/ml, with a total assay time of 4 h 30 min. Spike-and-recovery testing gave average recoveries of 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (range 81–100%, n=5) and 84% in heparin plasma (range 80–89%, n=5). Dilution linearity was demonstrated in serum, EDTA plasma and heparin plasma across dilutions of 1:2, 1:4, 1:8 and 1:16. Kit components include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A, Detection Solution B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4 °C or −20 °C as instructed.
