Human Thyroid Peroxidase ELISA Kit

Designed for the in vitro quantitative determination of human Thyroid Peroxidase (TPO) in tissue homogenates and other biological fluids, this sandwich ELISA kit from Antibodies.com employs a pre-coated 96-well microplate (12 x 8 well strips) with colorimetric detection. Captured TPO is bound by an immobilized antibody, then detected using a Biotinylated Anti-Thyroid Peroxidase/TPO Antibody followed by HRP-Avidin conjugate and TMB substrate; the resulting yellow product is read at 450 nm, with absorbance proportional to analyte concentration.

The assay has a working range of 0.312-20 ng/ml and a sensitivity of <0.112 ng/ml, with a total incubation time of 4h 30m. The kit is also validated for serum, EDTA plasma and heparin plasma in addition to tissue homogenates. Reported recovery values for neat samples are 80%-102% (serum, n=5), 81%-100% (EDTA plasma, n=5) and 80%-89% (heparin plasma, n=5), with mean recoveries of 91%, 90% and 84% respectively. Linearity on dilution at 1:2, 1:4, 1:8 and 1:16 spans 74%-108% across the matrices tested.

Reagent components include the pre-coated microplate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
