Human Thymus Activation Regulated Chemokine ELISA Kit

A sandwich ELISA (enzyme-linked immunosorbent assay) kit from Antibodies.com is designed for the in vitro quantitative determination of human TARC/CCL17 (Thymus Activation Regulated Chemokine). The assay is intended for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and has a reactivity profile limited to human samples.

The kit uses a pre-coated 96-well microplate (12 x 8 well strips) with an immobilised antibody specific for TARC/CCL17. Standards and samples are added, followed by a Biotinylated Anti-TARC/CCL17 Antibody and an HRP-Avidin conjugate; signal is developed with TMB substrate and read at 450 nm in a colorimetric microplate reader. The product range of the standard curve is 31.2–2000 pg/ml, with a stated sensitivity of 10.5 pg/ml (described as <10.5 pg/ml in the product information). Total assay time is 4 h 30 min, and components are stored at +4 °C or -20 °C as instructed.

Spike-and-recovery performance reported by the manufacturer for n=5 samples gave averages of 91% (serum, range 80%–102%), 90% (EDTA plasma, range 81%–100%) and 84% (heparin plasma, range 80%–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87%–107% (serum), 79%–108% (EDTA plasma) and 82%–105% (heparin plasma).
