Human Thymine DNA Glycosylase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, the Human TDG ELISA Kit is designed for the in vitro quantitative determination of human Thymine DNA Glycosylase (TDG) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate to which a TDG-specific antibody has been immobilised; standards and test samples are added, captured TDG is bound by a Biotinylated Anti-TDG Antibody, and detection is achieved with an HRP-Avidin conjugate. The enzymatic reaction is visualised using TMB substrate, producing a yellow product whose absorbance at 450 nm, read on a colorimetric microplate reader, is proportional to the TDG concentration present in the well and is quantified against the supplied standard curve.

Sensitivity is reported as <0.038 ng/ml with an assay range of 0.312–20 ng/ml, and the total assay time is approximately 4 hours 30 minutes. Reported mean recovery is 91% (range 80–102%) in serum, 90% (range 81–100%) in EDTA plasma, and 84% (range 80–89%) in heparin plasma (n=5 for each matrix). The assay is suitable for tissue homogenates, with kit components intended for storage at +4°C or –20°C as indicated on the labels; the kit must not be used past its stated expiration date.
