The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for THBS4 immobilized on the plate. Standards and samples are added, followed by a biotinylated anti-THBS4 antibody and HRP-avidin conjugate, and the TMB substrate produces a yellow product whose intensity is proportional to THBS4 concentration when read at 450 nm.

Reported sensitivity is 0.062 ng/ml with an assay range of 0.156-10 ng/ml, and the protocol takes 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the assay is performed on a colorimetric microplate reader.
