Human Thrombopoietin ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay kit from Antibodies.com is designed for the in vitro quantitative determination of human Thrombopoietin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs a sandwich enzyme immunoassay format using a 96-well microplate pre-coated with a Thrombopoietin-specific capture antibody. Standards and test samples are added to the wells, where Thrombopoietin is bound by the immobilized antibody. After washing, a Biotinylated Anti-Thrombopoietin Antibody is added, followed by an HRP-Avidin conjugate. Following further incubation and washing, TMB substrate solution is used to visualize the HRP enzymatic reaction, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of Thrombopoietin captured, and the concentration is calculated by reading the absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

Performance characteristics include a product range of 15.6-1000 pg/ml and a sensitivity of 6.1 pg/ml, with a total assay time of 4 hours 30 minutes. Spike-and-recovery values across sample types (n=5) were 80% - 102% for serum (average 91%), 81% - 100% for EDTA plasma (average 90%), and 80% - 89% for heparin plasma (average 84%). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
