Human Thioredoxin Reductase 1 ELISA Kit

From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of human Thioredoxin Reductase 1 (TXNRD1) in serum, plasma, tissue homogenates and other biological fluids, with reactivity specified for human samples.

The assay uses a 96-well microplate pre-coated with an antibody specific for TXNRD1. Standards and test samples are added to the wells, where TXNRD1 is captured by the immobilized antibody. After washing, a biotinylated anti-TXNRD1 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualize the reaction, producing a blue-coloured product that changes to yellow after addition of stop solution. Absorbance is read at 450 nm in a microplate reader and the TXNRD1 concentration is calculated from the standard curve. Total assay time is 4h 30m, detection is colorimetric and the platform is a pre-coated microplate configured as 12 x 8 well strips.

According to the manufacturer, the assay has a sensitivity of <0.123 ng/ml and a measuring range of 0.312–20 ng/ml. Reported recovery (n=5) spans 80–102% in serum (average 91%), 81–100% in EDTA plasma (average 90%) and 80–89% in heparin plasma (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported as 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components should be stored at +4°C or –20°C as instructed.
