Human TGF Beta Inducible Early Response Gene 1 ELISA Kit

A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human KLF10 in tissue homogenates, cell lysates, or other biological fluids. The assay uses an antibody specific for KLF10 pre-coated onto a 96-well microplate. Standards and samples are added to the wells and bound KLF10 is detected using a Biotinylated Anti-KLF10 Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue colour that changes to yellow on addition of the acidic stop solution; the yellow colour density is proportional to the amount of KLF10 captured in each well, and the concentration is calculated from the absorbance read at 450 nm in a microplate reader.

The assay has a reported sensitivity of <0.122 ng/ml and a standard range of 0.312–20 ng/ml, with a total incubation time of 4h 30m. Spiked recovery was evaluated in serum (n=5, range 80%–102%, average 91%), EDTA plasma (n=5, range 81%–100%, average 90%) and heparin plasma (n=5, range 80%–89%, average 84%). Linearity across 1:2 to 1:16 dilutions was demonstrated in serum, EDTA plasma and heparin plasma samples (n=5 each), with the reported ranges varying by dilution.

The kit is suitable for use with cell lysates and tissue homogenates. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used after the stated expiration date.
