Human Telomerase Reverse Transcriptase ELISA Kit

Offered by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Telomerase Reverse Transcriptase in tissue homogenates, cell lysates, and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, on which an antibody specific for Telomerase Reverse Transcriptase is immobilised.

In the workflow, standards and test samples are added to the wells, where the target is captured by the immobilised antibody. After washing, a biotinylated anti-Telomerase Reverse Transcriptase antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then applied and produces a blue-coloured product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the concentration is calculated by reference to a standard curve. Total assay time is 4 hours 30 minutes and detection is colorimetric.

Performance specifications include a product range of 15.6–1000 pg/ml and a sensitivity of <5.3 pg/ml. In spike-and-recovery testing (n=5), serum samples showed recovery of 80–102% (average 91%), EDTA plasma 81–100% (average 90%), and heparin plasma 80–89% (average 84%). Linearity of dilution was demonstrated across 1:2, 1:4, 1:8, and 1:16 dilutions for serum (74–107%), EDTA plasma (79–108%), and heparin plasma (82–105%). Kit components should be stored at +4°C or -20°C as instructed.
