Human T-Box Protein 3 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Tbx3 in tissue homogenates or other biological fluids. The assay uses the sandwich immunoassay technique: an antibody specific for Tbx3 is pre-coated onto the wells of a microplate, standards and test samples are added, and any Tbx3 present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Tbx3 Antibody is added followed by an HRP-Avidin conjugate. A TMB substrate solution visualises the HRP reaction as a blue product that turns yellow on addition of acidic stop solution, with yellow intensity proportional to the amount of Tbx3 captured. Concentration is calculated from the O.D. absorbance at 450 nm read on a microplate reader against a standard curve.

The reported sensitivity is <0.116 ng/ml and the assay range is 0.312–20 ng/ml. Detection is colourimetric, the platform is a pre-coated microplate supplied as 12 × 8 well strips, and the assay time is 4 h 30 min. Recovery has been evaluated in serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5), and linearity of dilution has been assessed at 1:2, 1:4, 1:8 and 1:16 across the same sample types. Components supplied include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with storage conditions ranging from +4 °C to −20 °C depending on the component.
