Human T-Box Protein 21 ELISA Kit

Sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human T-bet/Tbx21 in cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for T-bet/Tbx21 pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, followed by washing and incubation with a Biotinylated Anti-T-bet/Tbx21 Antibody, then HRP-Avidin conjugate. A TMB substrate produces a blue colour that changes to yellow upon addition of acidic stop solution; absorbance is read at 450nm in a microplate reader and the concentration is calculated from the standard curve.

Reported sensitivity is 0.052 ng/ml, with an assay range of 0.156–10 ng/ml and a total assay time of 4h 30m. Sample recovery data (n=5) show serum at 83–100% (average 91%), EDTA plasma at 90–103% (average 96%), and heparin plasma at 90–99% (average 94%). Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions: serum at 90–99%, 82–108%, 82–96%, 92–103%; EDTA plasma at 84–105%, 88–94%, 89–107%, 72–106%; heparin plasma at 89–92%, 88–108%, 84–102%, 90–98%. The kit includes a pre-coated microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, with storage at +4°C or -20°C depending on the component.
