Human Syntaxin 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Syntaxin 2 (STX2) in tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microtiter plate onto which an antibody specific for Syntaxin 2/STX2 has been immobilised. Standards and test samples are added to the wells and the Syntaxin 2/STX2 present in each sample is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Syntaxin 2/STX2 Antibody is applied and binds the captured analyte, after which an HRP-Avidin conjugate is added. The enzymatic reaction is then visualised using a TMB substrate solution that produces a blue-coloured product which changes to yellow upon addition of an acidic stop solution. The density of the yellow colour is proportional to the amount of Syntaxin 2/STX2 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a product range of 0.312-20 ng/ml and a sensitivity of <0.109 ng/ml, with a total assay time of 4h 30m and detection carried out on a colorimetric microplate reader. Sample recovery has been evaluated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5) and heparin plasma (range 80%-89%, average 84%, n=5), with dilution linearity values reported across 1:2, 1:4, 1:8 and 1:16 sample dilutions. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
