Human Syndecan 3 ELISA Kit

This Antibodies.com Human Syndecan 3 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of human Syndecan 3 in tissue homogenates or other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) with colorimetric detection, read on a microplate reader at 450 nm. The stated detection range is 0.625–40 ng/ml, with a sensitivity of <0.226 ng/ml, and the total assay time is 4 h 30 m. The procedure employs a biotinylated anti-Syndecan 3 detection antibody followed by an HRP–avidin conjugate, with signal generated by TMB substrate and visualised by an acidic stop solution; the resulting absorbance is proportional to the amount of Syndecan 3 captured in each well and is quantified against the standard curve.

Reported validation data for the kit give average recovery in serum of 91% (range 80–102%, n = 5), EDTA plasma 90% (range 81–100%, n = 5), and heparin plasma 84% (range 80–89%, n = 5). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries of 87–91%, 87–107%, 74–101% and 92–97% respectively for serum (n = 5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n = 5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n = 5). Kit components should be stored at +4 °C or –20 °C as instructed, and the kit should not be used past its expiration date.
