Human Synaptophysin ELISA Kit

This sandwich ELISA from Antibodies.com enables in vitro quantitative determination of human Synaptophysin in serum, plasma, tissue homogenates and other biological fluids. The assay employs a pre-coated 96-well microplate to which a capture antibody specific for Synaptophysin has been immobilised. Standards and samples are added to the wells; following incubation and washing, a biotinylated anti-Synaptophysin detection antibody is applied, followed by HRP-avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow upon addition of stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader.

Quantitative performance characteristics include a measuring range of 0.312–20 ng/ml and a sensitivity of <0.118 ng/ml. The total assay time is 4 hours 30 minutes. Spike-and-recovery testing across matrices yielded the following: serum (n=5) showed 80–102% recovery with an average of 91%, EDTA plasma (n=5) showed 81–100% with an average of 90%, and heparin plasma (n=5) showed 80–89% with an average of 84%. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16, with observed values across dilutions spanning 74–107% for serum, 79–108% for EDTA plasma, and 82–105% for heparin plasma.
