The assay uses an antibody specific for SMN/Gemin 1 pre-coated onto a 96-well microtiter plate. Standards and samples are added to the wells, the analyte is captured by the immobilized antibody, and detection is achieved using a biotinylated anti-SMN/Gemin 1 antibody followed by an HRP-avidin conjugate and TMB substrate. The reaction produces a blue product that turns yellow upon addition of stop solution, with absorbance measured at 450 nm on a colorimetric microplate reader and concentration calculated from a standard curve.

Sensitivity is <0.058 ng/ml with an assay range of 0.156-10 ng/ml, and the total assay time is 4h 30m. Spiking recovery averaged 91% in serum (range 80%-102%, n=5), 90% in EDTA plasma (range 81%-100%, n=5), and 84% in heparin plasma (range 80%-89%, n=5).
