Sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of human Surfactant Protein D/SP-D in serum, plasma, tissue homogenates, cell lysates, lung lavage fluid, cell culture supernates and other biological fluids. The assay employs the sandwich immunoassay principle: a capture antibody specific for SP-D is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added, captured SP-D is detected with a Biotinylated Anti-Surfactant Protein D/SP-D Antibody, and the signal is generated using HRP-Avidin conjugate with TMB substrate. The reaction is stopped with an acidic reagent and read at 450 nm on a colorimetric microplate reader, with concentration calculated from a standard curve.

Reported performance figures include a working range of 6.25–400 ng/ml and a sensitivity of <2.16 ng/ml, with a total assay time of 4 h 30 min. Spike-and-recovery data (n=5) average 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Linearity of dilution is also reported across serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions (n=5 per matrix), with values within ranges such as 74–107% across the matrices.

Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Components are to be stored at +4 °C or −20 °C as instructed, and the kit should not be used past its expiration date.
