Human Suppressors Of Cytokine Signaling 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Suppressors Of Cytokine Signaling 1 (SOCS1) in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips on which an antibody specific for SOCS1 has been immobilised. Standards and test samples are added, and the captured SOCS1 is detected sequentially with a Biotinylated Anti-SOCS1 Antibody and an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of an acidic stop solution. The intensity of the yellow colour is proportional to the amount of SOCS1 captured in each well, and the concentration is calculated by reading the absorbance at 450 nm on a colorimetric microplate reader and referring to a standard curve.

The assay has a reported sensitivity of <0.113 ng/ml and a product range of 0.312-20 ng/ml, with a total run time of 4h 30m. Spike-and-recovery performance has been characterised in serum (n=5, 80%-102%, average 91%), EDTA plasma (n=5, 81%-100%, average 90%) and heparin plasma (n=5, 80%-89%, average 84%). Dilution linearity has been evaluated at 1:2, 1:4, 1:8 and 1:16 across the same three sample matrices. Kit components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
