Description

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human Superoxide Dismutase 4/CCS in tissue homogenates, cell lysates or other biological fluids. The assay employs an antibody specific for Superoxide Dismutase 4/CCS pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where the analyte present is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Superoxide Dismutase 4/CCS Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Superoxide Dismutase 4/CCS captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The kit has a reported sensitivity of <0.117 ng/ml and a product range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Detection is colourimetric on a pre-coated microplate platform arranged as 12 x 8 well strips. Spike-and-recovery testing (n=5) reports the following average values: Serum 91% (range 80–102%), EDTA Plasma 90% (range 81–100%), and Heparin Plasma 84% (range 80–89%). Dilution linearity is provided for serum, EDTA plasma, and heparin plasma at 1:2, 1:4, 1:8, and 1:16 dilutions.

Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers. Components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
