Human Stratifin ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human stratifin (also known as 14-3-3 sigma/SFN) in serum, plasma, cerebrospinal fluid or other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for 14-3-3 sigma/SFN is pre-coated onto a microtiter plate; standards and test samples are added and the target in each sample is bound by the immobilized antibody. After washing, a Biotinylated Anti-14-3-3 sigma/SFN Antibody binds the captured target, followed by addition of an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration of 14-3-3 sigma/SFN is calculated against the standard curve.

Performance values quoted by the manufacturer include an assay range of 0.312–20 ng/ml and a sensitivity of <0.134 ng/ml. The assay is colourimetric, read on a microplate reader, with a total assay time of 4 hours 30 minutes. Kit components are stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date. Spiking recovery (n=5) is 80–102% (average 91%) in serum, 81–100% (average 90%) in EDTA plasma, and 80–89% (average 84%) in heparin plasma.
