A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Stearoyl Coenzyme A Desaturase (SCD1) in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) bearing an immobilised antibody specific for SCD1. Detection is achieved with a biotinylated anti-SCD1 antibody, an HRP-avidin conjugate and TMB substrate, and the enzymatic reaction is read at 450 nm on a colorimetric microplate reader.

Sensitivity is reported as <0.063 ng/ml, with a quantitative working range of 0.156–10 ng/ml and a total assay time of 4 hours 30 minutes. Recovery across five replicates averaged 91% in serum (range 80–102%), 90% in EDTA plasma (81–100%) and 84% in heparin plasma (80–89%). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16; in serum, recoveries spanned 87–91%, 87–107%, 74–101% and 92–97% respectively, in EDTA plasma 90–105%, 84–101%, 90–101% and 79–108%, and in heparin plasma 84–95%, 92–105%, 82–105% and 89–91% (n=5 each).

Kit components include a lyophilised standard (2 vials), detection solution A and detection solution B (120 µl each), 30× wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers (5 adhesive strips). Components are stored at +4 °C or −20 °C as instructed and the kit should not be used past its expiration date.
