Human Stathmin 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, intended for the in vitro quantitative determination of human Stathmin 1 in tissue homogenates, cell lysates or other biological fluids.

The kit uses a pre-coated 96-well microtitre plate (12 x 8 well strips) bearing an antibody specific for Stathmin 1. Standards and samples are added to the wells; Stathmin 1 is captured by the immobilized antibody and detected with a biotinylated anti-Stathmin 1 antibody followed by an HRP-avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of Stathmin 1 is calculated from the standard curve. Total assay time is 4 hours 30 minutes; sensitivity is less than 0.053 ng/ml and the assay range is 0.156–10 ng/ml.

Validation data supplied with the kit reports recovery in undiluted matrices (n=5): serum 80%–102% (average 91%), EDTA plasma 81%–100% (average 90%) and heparin plasma 80%–89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was measured in the same three matrices, with values within the ranges 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
