Human Spleen Tyrosine Kinase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, intended for the in vitro quantitative determination of human Spleen Tyrosine Kinase (Syk) in tissue homogenates, cell lysates and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for Syk. Standards and test samples are added to the wells, where Syk is captured by the immobilized antibody. After washing, a Biotinylated Anti-Syk Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that changes to yellow upon addition of an acidic stop solution; the absorbance of this yellow colour, read at 450 nm in a colorimetric microplate reader, is proportional to the amount of Syk captured in each well.

The kit shows a sensitivity of 0.244 ng/ml and an assay range of 0.625–40 ng/ml. Reported sample recovery values are 80–102% for serum (average 91%), 81–100% for EDTA plasma (average 90%) and 80–89% for heparin plasma (average 84%). Approximate total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or −20°C as instructed and the product should not be used past its expiration date.
