This Antibodies.com kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Somatostatin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay has a product range of 7.81-500 pg/ml and a sensitivity of <2.27 pg/ml, and is read on a colorimetric microplate reader at 450 nm. The pre-coated microplate platform supports the assay, which has a total run time of 4 hours 30 minutes.

The assay uses the sandwich enzyme immunoassay technique. An antibody specific for Somatostatin is pre-coated onto the plate; standards and samples are added and the Somatostatin present binds to the immobilized antibody. After washing, a Biotinylated Anti-Somatostatin Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of Somatostatin captured in each well.

Recovery was assessed in spiked samples: Serum (n=5) ranged 80% - 102% with an average of 91%; EDTA Plasma (n=5) ranged 81% - 100% with an average of 90%; Heparin Plasma (n=5) ranged 80% - 89% with an average of 84%. Linearity on dilution (1:2, 1:4, 1:8, 1:16) was 87-91%, 87-107%, 74-101% and 92-97% for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5). Kit components include a pre-coated microplate, lyophilized standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers, with storage at +4°C or -20°C as instructed. Do not use past expiration date. Sensitivity is given as <2.27 pg/ml.
