A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com, this Human Solute Carrier Family 1, Member 5 ELISA Kit is designed for the in vitro quantitative determination of human SLC1A5/ASCT2 in tissue homogenates, cell lysates, or other biological fluids. The assay has a reported range of 0.312–20 ng/ml and a sensitivity of <0.112 ng/ml. Detection is colourimetric and the assay is read on a microplate reader at 450 nm; total assay time is 4 hours 30 minutes.

The kit uses a pre-coated 96-well microplate arranged as 12 × 8 well strips. Standards and samples are added to the wells and the target is captured by the immobilized antibody. After washing, a biotinylated anti-SLC1A5/ASCT2 detection antibody is added, followed by HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, and the absorbance at 450 nm is used to calculate the SLC1A5/ASCT2 concentration from the standard curve. Reported sample recovery values are: serum 80–102% (average 91%, n=5), EDTA plasma 81–101% (average 91%, n=5), and heparin plasma 80–89% (average 84%, n=5), with dilution linearity data across 1:2, 1:4, 1:8, and 1:16 dilutions for all three sample types.

Kit components include a pre-coated microplate, lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and 5 adhesive plate sealers. Components are stored at +4°C or −20°C as instructed and the kit should not be used past its expiration date.
