Human Sirtuin 3 ELISA Kit

Antibodies.com supplies a sandwich ELISA kit for the in vitro quantitative determination of human SIRT3 (Sirtuin 3). The assay is intended for use with tissue homogenates, cell lysates and other biological fluids, and is reactive with human samples only. Detection is performed colorimetrically on a 96-well microplate pre-coated with an antibody specific for SIRT3, using a biotinylated anti-SIRT3 detection antibody and an HRP conjugate. Bound analyte is visualised with TMB substrate, producing a yellow product whose intensity is proportional to SIRT3 concentration and read at 450 nm against a standard curve.

The standard curve spans 0.312–20 ng/ml, with a stated sensitivity of <0.124 ng/ml, and the full assay is completed in 4h 30m. Spike-and-recovery was evaluated in serum (range 80–102%, average 91%), EDTA plasma (range 81–100%, average 90%) and heparin plasma (range 80–89%, average 84%), with linearity assessed at dilutions of 1:2, 1:4, 1:8 and 1:16. Kit components should be stored at +4°C or –20°C as instructed and the kit must not be used past its expiration date.
