The assay is specific to human STAT5B and is performed on a pre-coated microplate with a total run time of 4h 30m.

Quantification is achieved by reading absorbance at 450 nm on a colorimetric microplate reader; the intensity of the yellow colour developed after addition of TMB substrate and acidic stop solution is directly proportional to the amount of STAT5B captured in each well and is calculated against the supplied standard curve. The kit has a reported sensitivity of <0.137 ng/ml and a measuring range of 0.312–20 ng/ml.
