The Antibodies.com STAT5A ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Signal Transducer and Activator of Transcription 5A (STAT5A) in tissue homogenates, cell lysates, and other biological fluids. The kit is also suitable for use with serum, EDTA plasma, and heparin plasma samples and is configured for colorimetric detection on a microplate reader.

The assay employs the sandwich enzyme immunoassay technique using a pre-coated microplate. An antibody specific for STAT5A is pre-coated onto the plate; standards and test samples are added and STAT5A present in each sample binds to the immobilised antibody. After incubation and washing, a Biotinylated Anti-STAT5A Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate is then added to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of STAT5A captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against the standard curve.

Reported performance figures include a sensitivity of <0.112 ng/ml and an assay range of 0.312–20 ng/ml. Average recovery values are 91% for serum (range 83–100%, n=5), 95% for EDTA plasma (range 90–100%, n=5), and 96% for heparin plasma (range 91–101%, n=5). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma. The total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
