This Antibodies.com kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of human STAT4 (Signal Transducer and Activator of Transcription 4) in tissue homogenates, cell lysates or other biological fluids.

The assay uses the sandwich enzyme immunoassay technique, in which an antibody specific for STAT4 is pre-coated onto a microplate. Standards and test samples are added, and captured STAT4 is detected with a Biotinylated Anti-STAT4 Antibody followed by an HRP-Avidin conjugate. A TMB substrate solution is used to visualise the reaction, producing a blue product that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and STAT4 concentration is calculated by reference to a standard curve.

The reported assay range is 0.312–20 ng/ml with a sensitivity of 0.121 ng/ml, and total assay time is 4 hours 30 minutes. Average recovery values are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5), and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity is provided for serum, EDTA plasma, and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16. Kit components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
