The assay uses a microplate pre-coated with an antibody specific for STAT2, paired with a biotinylated anti-STAT2 detection antibody and an HRP-avidin conjugate. Captured STAT2 is visualised using a TMB substrate, producing a colour reaction whose intensity, read at 450 nm on a colorimetric microplate reader, is proportional to the amount of STAT2 present.

According to the manufacturer, the kit has a sensitivity of 0.108 ng/ml, a standard curve range of 0.312–20 ng/ml, and a total assay time of 4 hours 30 minutes.
