Human Serum Amyloid A2 ELISA Kit

The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips and is read on a colorimetric microplate reader at 450 nm.

The reported product range is 0.625–40 ng/ml with a sensitivity of 0.277 ng/ml. The workflow follows a standard sandwich immunoassay: standards and samples are added to the pre-coated wells and the target is captured by the immobilized antibody; a Biotinylated Anti-Serum Amyloid A2 Antibody is then added, followed by an HRP-Avidin conjugate and TMB substrate. Catalysis yields a blue product that turns yellow on addition of acidic stop solution, with the density of yellow proportional to the amount of Serum Amyloid A2 captured. Total assay time is 4 h 30 m, and kit components are stored at +4 °C or −20 °C as instructed.

The kit has been validated in serum, EDTA plasma and heparin plasma. Across the three sample types (n = 5 each), average recovery was 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 gave recovery ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
