Human Serine Peptidase Inhibitor Kazal Type 5 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of human SPINK5/LEKTI (Serine Peptidase Inhibitor Kazal Type 5) in tissue homogenates, cell lysates or other biological fluids. The kit is reactive with human samples and is read on a colorimetric microplate reader at 450 nm.

Reported performance characteristics include a sensitivity of 0.116 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Average recovery across serum samples (n=5) was 91% (range 80%–102%), 91% in EDTA plasma (n=5, range 81%–101%) and 84% in heparin plasma (n=5, range 80%–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is reported for serum, EDTA plasma and heparin plasma samples.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for SPINK5/LEKTI is immobilised on the plate; standards and test samples are added, followed by a biotinylated anti-SPINK5/LEKTI antibody and an HRP-avidin conjugate. TMB substrate is used to visualise the reaction, producing a blue product that turns yellow upon addition of stop solution; the yellow intensity is proportional to SPINK5/LEKTI concentration. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
