Human Septin 3 ELISA Kit

This Antibodies.com sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Septin 3 in tissue homogenates and other biological fluids, including serum, EDTA plasma, heparin plasma, cell culture supernatant, tissue, and cell lysates. The assay uses a 96-well microplate pre-coated with an antibody specific for Septin 3. Standards and samples are added to the wells, followed by incubation with a biotinylated anti-Septin 3 antibody and an HRP-avidin conjugate. Detection is colourimetric, using TMB substrate and reading absorbance at 450 nm on a microplate reader.

The standard curve covers 0.312–20 ng/ml with a sensitivity of 0.123 ng/ml. Reported average recovery in spiked samples is 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5), and 84% for heparin plasma (range 80–89%, n=5). Linearity across 1:2, 1:4, 1:8, and 1:16 dilutions is 87–91%, 87–107%, 74–101%, and 92–97% for serum; 90–105%, 84–101%, 90–101%, and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% for heparin plasma. Total assay time is 4 hours 30 minutes. The kit includes a pre-coated microplate in a 12 x 8 well strip format, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with components stored at +4°C or −20°C as instructed.
