Human Semaphorin 4B ELISA Kit

From Antibodies.com, this Human Semaphorin 4B ELISA Kit is intended for the in vitro quantitative determination of human Semaphorin 4B (referred to as SEMAC in the manufacturer's literature) in tissue homogenates or other biological fluids. The kit uses a sandwich enzyme immunoassay format on a pre-coated 96-well microplate arranged as 12 x 8 well strips, is reactive with human samples, and is read on a colourimetric microplate reader at 450 nm. Reported sensitivity is 0.071 ng/ml and the assay range is 0.156-10 ng/ml, with a total assay time of 4h 30m.

The protocol uses a biotinylated anti-SEMAC detection antibody followed by an HRP-avidin conjugate and TMB substrate; an acidic stop solution yields a yellow product whose absorbance is proportional to bound SEMAC and is interpolated against a standard curve. Average recoveries stated by the manufacturer are Serum 91% (range 83-100%, n=5), EDTA Plasma 95% (range 91-99%, n=5) and Heparin Plasma 94% (range 90-99%, n=5), with dilution linearity data provided for 1:2, 1:4, 1:8 and 1:16 sample dilutions across the same three matrices.
