Human Selectin, Platelet ELISA Kit

Sandwich-format enzyme-linked immunosorbent assay (ELISA) marketed under the Antibodies.com brand, intended for the in vitro quantitative determination of human CD62P (Selectin, Platelet) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique with an antibody specific for CD62P pre-coated onto a 96-well microplate (12 x 8 well strips). Standards and test samples are added to the wells and captured by the immobilised antibody. After washing, a Biotinylated Anti-CD62P Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue-coloured product that converts to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader, and CD62P concentration is calculated against the standard curve.

The quantitative assay covers a measuring range of 78.1–5000 pg/ml with a sensitivity of <31 pg/ml. Total assay time is 4 h 30 min. Kit components are stored at +4 °C or −20 °C as instructed and must not be used past the expiration date.

Reported average recovery from undiluted samples (n=5) is 91% for serum, 90% for EDTA plasma and 84% for heparin plasma. Dilution linearity has been evaluated for serum, EDTA plasma and heparin plasma across dilutions of 1:2, 1:4, 1:8 and 1:16.
