Human Secretagogin ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of human SCGN/Secretagogin in serum, plasma, tissue homogenates or other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for SCGN/Secretagogin immobilised on the plate to capture the analyte from standards and test samples. A Biotinylated Anti-SCGN/Secretagogin Antibody is then added, followed by an HRP-Avidin conjugate, and TMB substrate is used to visualise the enzymatic reaction by catalysis to produce a blue-coloured product that becomes yellow after addition of acidic stop solution. The colour density is read by measuring O.D. absorbance at 450 nm in a microplate reader and the analyte concentration is calculated from the standard curve.

The assay range is 78.1-5000 pg/ml with sensitivity of <29 pg/ml. Total assay time is 4h 30m and detection is colourmetric. Reported average recovery is 91% (range 80%-102%) in serum (n=5), 90% (range 81%-100%) in EDTA plasma (n=5) and 84% (range 80%-89%) in heparin plasma (n=5). Linearity has been reported across dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma (n=5).

The kit contains a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120μl), Detection Solution B (120µl), Wash Buffer 30X (20ml), Sample Dilution Buffer (45ml), TMB Substrate (9ml), Stop Solution (6ml) and 5 adhesive plate sealers. Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.
