Human Secondary Lymphoid Tissue Chemokine ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of human CCL21 (Secondary Lymphoid Tissue Chemokine) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The immunoassay uses a pre-coated microplate (12 × 8 well strips) bearing an immobilised antibody specific for CCL21. Standards and samples are incubated in the wells, after which a biotinylated anti-CCL21 detection antibody is added to form a sandwich complex. Following incubation with an HRP-avidin conjugate and washing, TMB substrate is added; the resulting colour change is read at 450 nm on a colorimetric microplate reader and the CCL21 concentration is calculated against the standard curve.

The working assay range is 31.2–2000 pg/ml with a sensitivity of 12.3 pg/ml, and a typical run takes 4 hours 30 minutes. Spike-and-recovery performance has been characterised in serum (range 80%–102%, average 91%), EDTA plasma (81%–100%, average 90%) and heparin plasma (80%–89%, average 84%), with linearity demonstrated for serum, EDTA plasma and heparin plasma across sample dilutions from 1:2 to 1:16. Kit components are supplied for storage at +4°C or –20°C as indicated in the manual, and the reagents should not be used past their stated expiration date.
