Human SATB Homeobox Protein 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human SATB Homeobox Protein 1 in tissue homogenates and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for SATB1, with detection via a biotinylated anti-SATB1 antibody and HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm. The stated assay range is 0.312–20 ng/ml with a sensitivity of 0.102 ng/ml, and the total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed.

Manufacturer-reported recovery values (n=5) include serum at an average of 91% (range 80–102%), EDTA plasma at 90% (range 81–100%), and heparin plasma at 84% (range 80–89%). Linearity has been evaluated at dilutions of 1:2, 1:4, 1:8 and 1:16 across the same matrices. Required materials not supplied with the kit include a microplate reader capable of measuring absorbance at 450 nm ± 10 nm, a 37°C incubator, multi- and single-channel pipettes, and deionised or distilled water. Sample types covered by the protocol include serum, EDTA or heparin plasma, cell culture supernatants, tissue homogenates, cell lysates and other biological fluids, with aliquoted storage recommended at -20°C or -80°C to limit freeze/thaw cycles.
