An enzyme-linked immunosorbent assay from Antibodies.com enables the in vitro quantitative determination of human Sarcolipin (SLN) in tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for SLN pre-coated onto a microplate. Standards and test samples are added to the wells, where SLN is captured by the immobilized antibody. After washing, a Biotinylated Anti-SLN Antibody binds the captured SLN, followed by incubation with HRP-Avidin conjugate. TMB substrate is then added, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The concentration of SLN is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and comparing to a standard curve.

Key performance characteristics include a sensitivity of 0.121 ng/ml and an assay range of 0.312-20 ng/ml. The assay uses a colorimetric detection method with a total assay time of 4h 30m.

The kit has been validated for use with serum, EDTA plasma, and heparin plasma samples. In undiluted samples, average recovery rates of 91% (serum, n=5, range 80–102%), 90% (EDTA plasma, n=5, range 81–100%), and 84% (heparin plasma, n=5, range 80–89%) have been reported. Dilution linearity has been demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across all three sample types. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
