Human SAKL ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human Soluble alpha Klotho in serum, plasma, tissue homogenates, and other biological fluids. It is specific for Soluble alpha Klotho with no reported cross-reactivity with other analogs or variants.

The assay runs in 4 hours on a pre-coated microplate. Standards and samples are added to the wells where the target binds to the immobilized capture antibody; a biotinylated detection antibody then forms an antibody–antigen–antibody sandwich complex. After incubation and washing, HRP-Streptavidin conjugate and TMB substrate are added, producing a blue product that turns yellow on addition of stop solution. Absorbance is read at 450 nm and concentration is calculated against a standard curve spanning 93.75–6000 pg/mL. Reported sensitivity is <56.25 pg/mL.

Average recovery was 98% for serum (range 87–104%), 92% for EDTA plasma (88–96%), and 97% for heparin plasma (90–104%), with dilution linearity assessed at 1:2, 1:4 and 1:8. Kit contents include a pre-coated microplate, lyophilized standard, sample dilution buffer, biotinylated detection antibody, antibody dilution buffer, HRP-streptavidin conjugate, SABC dilution buffer, TMB substrate, stop solution, wash buffer, plate sealers and a foil zip-sealed pouch. Components are shipped at +4°C and stored at +4°C or −20°C as directed in the protocol manual.
